Indications

Temodar is a cancer medication used in combination with radiation to treat certain types of brain tumours.

Temozolomide is an alkylating drug that affects cell DNA to inhibit the growth and spread of cancer cells throughout your body. This activity helps Temodar treat certain types of brain tumours caused by cancerous cells alongside radiation therapy after other medications had no successful response.

Dosage and Administration

The dose of Temodar that is prescribed will vary depending on the individual being treated. Follow all your healthcare provider’s instructions and read packaging labels carefully to ensure the best results during treatment.

Do not take more Temodar than prescribed.

  • Swallow capsules whole, do not crush or chew
  • Take at the same time each day
  • Can be taken with or without food but take doses the same way each day
  • If you experience nausea or vomiting, take Temodar on an empty stomach or before bed
  • Store at room temperature away from children and pets

To treat glioblastoma:

  • Most adults start by taking 75mg once a day for 42 days with radiation therapy
  • 4 weeks following this 42-day treatment, adults should take Temodar once daily for days 1 to 5 of 28-day cycles for 6 cycles
  • Most adults take 150mg once daily for five days for the first cycle before increasing to 200mg if needed starting with the second cycle
  • Do not increase your dose for cycles 3 and 4 if you did not initiate 200mg starting with cycle 2

To treat refractory anaplastic astrocytoma:

  • Most adults take 150mg once a day for the first 5 days of each 28-day cycle
  • Doses may increase to 200mg for each of the five days of the next cycle
  • Treatment should continue until your cancer improves or for a maximum of 2 years

Side Effects

Temodar may cause some side effects to develop while affecting your cells. Speak with your healthcare provider to learn more about any symptoms that you may experience during treatment.

Common Temodar side effects:

  • Nausea, vomiting, diarrhea, constipation
  • Headache, tiredness
  • Seizure
  • Decreased appetite
  • Hair loss

Serious Side Effects

Some more serious adverse reactions may occur during treatment with Temodar. Contact your healthcare provider right away if you notice any of the following symptoms:

  • Seizure
  • Sudden chest pain or discomfort, wheezing, dry cough
  • Low blood cell counts (fever, chills, mouth sores, skin sores, easy bruising, unusual bleeding, pale skin, cold hands and feet, light-headedness, shortness of breath)
  • Liver problems (nausea, upper stomach pain, itching, tiredness, loss of appetite, dark urine, clay-coloured stools, jaundice)

Allergic Reaction

Do not take Temodar if you are allergic to temozolomide or any of the other capsule ingredients. If you notice any signs of an allergic reaction, stop taking this medication and call your healthcare provider.

Temodar allergy symptoms may include:

  • Hives
  • Difficulty breathing
  • Swelling of face, lips, tongue, or throat

Cautions

Do not take Temodar if you are pregnant because the medication can harm your unborn baby.

Temodar can affect the development of an unborn baby if either the mother or father is taking the medication. Use effective birth control during treatment and for 6 months after your final dose if you are female or for at least 3 months if you are male.

This medication may make it more difficult for a man to impregnate someone during treatment. Ensure that you still use effective birth control to prevent pregnancy while taking Temodar because it can harm an unborn baby.

To ensure your safety when taking Temodar, tell your healthcare provider if you have ever had any of the following:

  • Liver disease
  • Kidney disease

Temodar therapy may increase your risk of developing certain types of bone marrow cancer. Speak with your healthcare provider about your risks.

Do not breastfeed during treatment with Temodar and for at least 1 week following your last dose.

Some drugs may interact with Temodar and cause adverse reactions. Tell your healthcare provider about all the medications you take, including over the counter medicines, vitamins, and herbal products.
